Tetrabutylammonium phosphate (TBAP) is positively charged and can interact with negatively charged DNA through electrostatic interaction to improve the resolution of DNA fragments. In addition, TBAP has no absorbance at 260 nm, which does not affect the detection of DNA.
TBAP Assisted DNA Fragment Isolation
As an ion pair reagent, TBAP is used as an additive in the background solution to improve the separation of DNA fragments, because positively charged TBA can interact with negatively charged DNA through electrostatic interaction. In addition, TBA has no absorbance at 260 nm, which does not affect the detection of DNA. The concentration of TBAP in background solution was optimized. When the concentration of TBAP was 15 mM, the resolution of DNA fragments was improved.
The reason for the improvement of DNA separation by adding TBAP was studied. The ultraviolet spectra of DNA with and without TBAP were detected. When TBAP is added to the background solution, the maximum absorption wavelength of DNA has a slight red shift, and the absorbance near 310 nm also has a slight red shift. In addition, after adding TBAP, the absorbance of DNA at 260 nm is higher than that without TBAP. The results show that under the experimental pH = 7.3, TBA has positive charge, DNA has negative charge, and TBAP has electrostatic interaction with DNA. In addition, the detection sensitivity of DNA fragments has been improved.
